Evaluation on the Pathogenesis of Streptococcus agalactiae in Nile Tilapia (Oreochromis niloticus) BABT
Pretto-Giordano,Lucienne Garcia; Müller,Ernst Eckehardt; Freitas,Julio Cesar de; Silva,Vanessa Gomes da.
The pathogenesis of a Streptococcus agalactiae was evaluated in a three-period experiment. Two groups of 40 fishes were intraperitoneally (i.p.) challenged in each experimental period with different infective doses of the pathogen. Doses varied from 1.0 x 10(6) to 1.5 x 10(8) CFU/fish. One group of 40 tilapia i.p. injected with tryptic soy broth (TSB) was used as a control group in each period. Mortalities varied from 67.5% in group 8 (infective dose 1.0 x 10(6) CFU/fish) to 90.0% in group 1 (infective dose 1.5 x 10(8) CFU/fish). Significant differences in mortalities were found only between group 8 and each of the other groups, except group 5 (infective dose 6.0 x 10(6) CFU/fish; mortality 75.0%). The highest mortality coefficients were observed in days...

First record of Borrelia burgdorferi B31 strain in Dermacentor nitens ticks in the northern region of Parana (Brazil) BJM
Gonçalves,Daniela Dib; Carreira,Teresa; Nunes,Mónica; Benitez,Aline; Lopes-Mori,Fabiana Maria Ruiz; Vidotto,Odilon; Freitas,Julio Cesar de; Vieira,Maria Luísa.
The aim of this study was to investigate the presence of DNA of Borrelia burgdorferi sensu lato (s.l.) in ticks that feed on horses used for animal traction in rural Jataizinho, Parana, Brazil. Between February and June 2008, a total of 224 ticks was collected of which 75% were identified as Dermacentor nitens and 25% as Amblyomma cajenense. To amplify B. burgdorferi s.l. DNA, the intergenic space region (ISR) between the 5S (rrf) 23S (rrl) rRNA genes was used as targets for nested-PCR. Two ticks of the D. nitens species were positive for B. burgdorferi s.l. Both species showed a fragment of 184 bp, but the sequencing revealed 99.9% homology with the B. burgdorferi sensu stricto (s.s.) strain B31. These results showed, for the first time, the presence of...

Investigation of the possible role of Chlamydophila abortus in reproductive failures in nrazilian herds of domestic ruminants BABT
Silva-Zacarias,Francielle Gibson da; Alfieri,Amauri Alcindo; Spohr,Kledir Anderson Hofstaetter; Lima,Bruna Azevedo de Carvalho; Oliveira,Rosângela Claret de; Turilli,Carlo; Lunardi,Michele; Otonel,Rodrigo Alejandro Arellano; Freitas,Julio Cesar de.
Chlamydophila abortus (C. abortus) infection is related to reproductive failure in domestic ruminants. Although it has not been well characterized worldwide, this pathogen has already been identified in some European countries and in the USA. In Brazil, preliminary studies have shown serological evidence of C. abortus infection in herds with low antibody prevalence. Until now, the identification of C. abortus in biological samples from females presenting reproductive failures has not been described in Brazilian herds of domestic ruminants. The aim of this study was to evaluate the presence of the C. abortus in a collection of abortions from cattle (n=85), sheep (n=12), and goats (n=8), in samples of vaginal mucus from cows (n=13), sheep (n=90), and goats...

Isolation of leptospira Serovars Canicola and Copenhageni from cattle urine in the state of Paraná, Brazil BJM
Zacarias,Francielle Gibson da Silva; Vasconcellos,Silvio Arruda; Anzai,Eleine Kuroki; Giraldi,Nilson; Freitas,Julio Cesar de; Hartskeerl,Rudy.
In 2001, 698 urine samples were randomly collected from cattle at a slaughterhouse in the State of Paraná, Brazil. Direct examination using dark field microscopy was carried out immediately after collection. Five putative positive samples were cultured in modified EMJH medium, yielding two positive cultures (LO-14 and LO-10). Typing with monoclonal antibodies revealed that the two isolates were similar to Canicola (LO-14) and Copenhageni (LO-10). Microscopic agglutination test results show that Hardjo is the most common serovar in cattle in Brazil. Rats and dogs are the common maintenance hosts of serovars Copenhageni and Canicola. The excretion of highly pathogenic serovars such as Copenhageni and Canicola by cattle can represent an increasing risk for...

Dreer,Márcia Küster de Paula; Gonçalves,Daniela Dib; Caetano,Isabel Cristina da Silva; Gerônimo,Edson; Menegas,Paulo Henrique; Bergo,Danilo; Lopes-Mori,Fabiana Maria Ruiz; Benitez,Aline; Freitas,Julio Cesar de; Evers,Fernanda; Navarro,Italmar Teodorico; Martins,Lisiane de Almeida.
Background : Leptospirosis, toxoplasmosis and brucellosis are diseases with worldwide distribution. Among stray dogs, these zoonoses are facilitated by direct contact with other animal species, by the habit of scavenging garbage and hunting in search of food, drinking standing water, smelling other animals' urine, licking female genitalia and the sexual act itself. The objective of this study was to detect antibodies anti-Toxoplasma gondii, anti-Leptospira spp., anti-Brucella canis and anti-Brucella abortus in stray dogs housed in shelters at Umuarama city, Paraná, Brazil. In order to detect toxoplasmosis, indirect immunofluorescence assay (IFA) was performed, agglutination microscopic (MAT) test for leptospirosis and agar gel immunodiffusion (AGID) and...

Validation of a PCR Assay for Chlamydophila abortus rRNA gene detection in a murine model BABT
Silva-Zacarias,Francielle Gibson da; Alfieri,Amauri Alcindo; Spohr,Kledir Anderson Hofstaetter; Lima,Bruna Azevedo de Carvalho; Negrão,Fábio Juliano; Lunardi,Michele; Freitas,Julio Cesar de.
Chlamydophila abortus (C. abortus) is associated with reproductive problems in cattle, sheep, and goats. Diagnosis of C. abortus using embryonated chicken eggs or immortalized cell lines has a very low sensitivity. Polymerase chain reaction (PCR) assays have been used to detect C. abortus infection in clinical specimens and organ fragments, such as placenta, fetal organs, vaginal secretions, and semen. The aim of this study was to develop a PCR assay for the amplification of an 856-bp fragment of the rRNA gene of the Chlamydiaceae family. The PCR assay was evaluated using organs from 15 mice experimentally infected with the S26/3 reference strain of C. abortus. The results of the rRNA PCR were compared to the results from another PCR system (Omp2 PCR) that...
